Peroxisome proliferator activated receptors and your ticker

Everyone knows by now that having diabetes raises the risk for heart disease in various forms. Wouldnt it be interesting to understand the biological reasons for this connection? Well, it turns out that various factors seem to be relevant, and one of them involves a variety of paths that all pass through the territory of a rather interesting protein called PPAR-γ.

Not only is PPAR-γ implicated in processes related to both diabetes and heart disease, but it turns out that some drugs used to control diabetes also affect the risk of heart disease – because of PPAR-γ.

Well begin the discussion by looking at recent research on how PPAR-γ affects heart function.

First, of course, we should explain what a peroxisome proliferator-activated receptor is. The name is somewhat off-putting, and nowadays most people just write PPAR. Its also a bit of a historical artifact, in that this class of proteins was first investigated in connections with peroxisomes, which are cellular organelles that participate in the metabolism of fatty acids. Thats an important clue right there, because if we are dealing with fat metabolism, there may well be connections with conditions such as obesity and cardiovascular disease.

It turns out that is only a part of what PPARs are connected with.

A PPAR is not a cell surface receptor. Instead it is a nuclear receptor, meaning that its a protein found in the interior of cells that (like a surface receptor) is activated when it connects with hormones and similar molecules. Such molecules that bind to a receptor are called ligands.

Initially, the ligands in question were known to cause proliferation of peroxisomes, but now many other kinds of ligands that activate PPARs have been identified.

Once a PPAR has been activated it can affect the expression of many different genes, because it acts as a transcription factor.

Three important PPARs are known: PPAR-α, PPAR-β (also called PPAR-δ), and PPAR-γ. Its the last of these well be concerned with here. In fact, PPAR-γ seems to affect many cellular processes related to metabolism and other things. Recent research that we may discuss another time (see here) has shown that there are about 5300 sites in the DNA of a fat cell that PPAR-γ can bind to, and hence potentially affect the expression of nearby genes. So its not surprising that PPAR-γ is involved in quite a lot of cellular business.

Incidentally, all three PPARs are produced from the same gene, with the variant forms being due to alternative splicing.

The research we want to highlight here deals with how PPAR-γ is linked with the daily rise and fall of heart rate and blood pressure. Such things that are part of the normal circadian rhythm, in animals, are usually regulated from the central nervous system. But that doesnt seem to be the only regulator:

What Makes The Heart Tick-tock (12/2/08)
Researchers have new evidence to show that the heart beats to its own drummer, according to a report in the December issue of the journal Cell Metabolism. Theyve uncovered some of the molecular circuitry within the cardiovascular system itself that controls the daily rise and fall of blood pressure and heart rate. The findings might also explain why commonly used diabetes drugs come with cardiovascular benefits, according to the researchers.

"This is the first study to demonstrate that a peripheral clock plays a role in the circadian rhythm of blood pressure and heart rate," said Tianxin Yang of the University of Utah and Salt Lake Veterans Affairs Medical Center.

While much progress has been made over the years in understanding the bodys master clock in the brain, the new study offers one of the first glimpses into the biological function of peripheral clocks in maintaining the circadian rhythms of tissues throughout the body, the researchers said.

There has already been reason to suspect that PPAR-γ is involved in this:
Earlier studies suggested a role for the nuclear receptor called peroxisome proliferator-activated receptor-γ (PPAR-γ) in clock function. PPAR-γ is perhaps best known as the molecular target for a class of widely prescribed and effective diabetes drugs called thiazolidinediones (TZDs), including rosiglitazone (trade name Avandia) and pioglitazone (trade name Actos). Those diabetes drugs are known to come with a side benefit: they have protective effects on the cardiovascular system.

The new research shows that the circadian variation in heart rate and blood pressure is disrupted simply by eliminating PPAR-γ from cardiovascular cells. The elimination was effected by working with two strains of mice in which suitable genes had been knocked out:
The researchers found that both knockout strains showed a significant reduction of circadian variations in blood pressure and heart rate. .... The mice also showed declines in variation of norepinephrine/epinephrine in their urine—a measure of activity of the sympathetic nervous system, which plays a key role in heart rate and blood pressure.

The animals had impairments in the rhythmicity of the major clock genes, including Bmal1, a transcription factor that controls the activity of other core clock components, they report. By treating the mice with the diabetes drug rosiglitazone, they were able to increase the activity of Bmal1 in the animals aortas, the largest artery of the body that issues blood from the heart, and further study showed that the core clock gene is directly controlled by PPAR-γ.

What, more precisely, is the role of PPAR-γ in affecting rhythmicity? Apparently the effect is indirect, due to its abillity to activate Bmal1, which is known to be an important clock protein. This is indicated because rosiglitazone seems to be able to compensate for missing PPAR-γ.

Interestingly, other recent research has shown that the sirtuin protein SIRT1 also affects Bmal1. (See here.) this may be significant, since SIRT1 has gene-silencing effects that depend on nutritional factors.

What other processes is PPAR-γ involved with? Better-known than its effect on cardiovascular circadian rhythm is its role in fatty acid storage and glucose metabolism, and hence its connection with diabetes. But well have to look at that another time.

Skin Care My Best Tips to Make Your Skin Look Younger

Skin Care: My Best Tips to Make Your Skin Look Younger
The right (& wrong) ways to cleanse, moisturize & exfoliate your skin

To keep your face looking as youthful as possible, you need to properly cleanse, exfoliate and moisturize your skin while never leaving the house without at least 30 SPF sunscreen. Ive dubbed this process "The Basic 4-Step Skincare Regimen," which I outline in this article.

skincare-am.com

But before we go there, how you care for your skin is utterly dependent on the type of skin you have: oily, normal/combination, dry, sensitive or sun- damaged. Find out what skin type you have.

The Basic 4-Step Skincare Regimen Step 1: Cleansing

Simple is key here. You need to find a good cleanser that your skin responds well to, and stick with it: See the best cleansers for your skin type.
You can find a good cleanser at the drugstore. Theres no need to spend $40 on a fancy wash. Avoid bar soaps as they tend to dry out the skin. According to Rona Berg, in her book, "Beauty," a French cosmetics executive once told her, "Soap should only ever touch your skin from the neck down." I agree. Choose a creamy cleanser if you have dry skin or a clear cleanser if you have oily skin.

Be careful not to cleanse too often, you risk over-cleansing skin, see signs you are overcleansing your skin. You really only need to wash your face at night to remove makeup and sunscreen, which can clog pores. If you have dry skin, consider cold cream like Ponds, which the French use. Simply apply cream, then wipe off, no water needed (if you have hard water, it can be especially harsh on skin). Most women prefer the water method: Use warm water to loosen dirt and clogged pores. Use a dime-sized bit of cleanser, then rinse with cool or lukewarm water. I personally swear by my Clarisonic Mia, as does pretty much everyone I know who has one.

Make sure to remove eye makeup with a proper makeup remover. The area around the eye is delicate so dont pull or rub too hard.

In the morning, a splash of lukewarm water is all you need (I find its great for removing excess oils from your nightly moisturizing). Never wash your face with hot or cold water (both can cause broken capillaries).

Step 2: Exfoliate

Exfoliation is the step most people skip in their weekly skincare routine. But trust me, if you start properly exfoliating your skin, you will notice an almost immediate difference. According to Berg, one of the reasons mens skin looks more youthful than womens is because men tend to exfoliate daily when they shave.
I use a facial scrub weekly to exfoliate my skin, but I also find using a washcloth when I cleanse my skin works just as well. I simply put a dab of cleanser on a damp washcloth and massage the cleanser into my skin in a circular motion. After a quick rinse, any sign of dead skin is erased. You can also exfoliate skin via microdermabrasion, chemical peels and retinoids. In my article, How to Exfoliate, I share all my tips and tricks to proper exfoliation. Including why you should throw out the loofah.

See my list of the best facial scrubs and microdermabrasion kits.

Scrubs work by removing the top layer of dead skin cells that tend to dull your complexion. We find exfoliating skin once a week with a microdermabrasion kit keeps skin glowing year-round. Make sure you use a gentle scrub with tiny grains. Big grains in cheap scrubs can tear skin and cause more harm than good. My favorite microdermabrasion kit is made by Lancome.

In the hour it takes to get a chemical peel, you can take off five years from your face. Cant afford the price tag for a monthly peel? Try some over-the-counter peels that work over the course of a month. I prefer MD Skincares.

Retinoids (such as Retin-A or the more moisturizing Renova) also work by removing the top layer of dead skin cells while also generating collagen in the skin. "Collagen is the skins structural fiber," dermatologist Dennis Gross said in O Magazine. "As we get older, it breaks down, creating lines and large pores." Skincare experts disagree on all sorts of things, but most of them consider retinoids to be a miracle skin saver. Im addicted to Retin-A, which I pick up in Mexico on my yearly jaunts.

Should you use a toner? Some people swear by toners, but many beauty experts do not (I once read a skincare expert claim, "toners are only for copy machines"). Toners are meant to remove all remaining traces of oil, makeup and dirt, but a good cleanser should do this. I firmly believe its up to you. If you like the way your skin feels with a toner. Buy it. Use it. Enjoy it.

Step 3: Moisturize

While I know of at least one famous beauty editor who swore skin doesnt need moisturizer, basically everyone else Ive read disagrees and is an adamant believer in it. A basic rule of beauty is that if you have dry skin, you should invest in a basic moisturizer. So how much should you moisturize? Your skin will tell you. When your skin is tight, its crying out for moisture. Be careful not to over-moisturize -- this can clog pores.
See my list of the best moisturizers on the market.

Are eye creams necessary? Well maybe. Some beauty experts strongly recommend eye creams. Why? The skin around the eye contains no fatty tissue and is therefore very thin and susceptible to wrinkles. Special eye creams are formulated to "thicken" this area. Yet other experts (including the beauty editors of Allure in their new book) claim your daily lotion works around the eyes just as well.

See my list of the best eye creams.

Step 4: Apply Sunscreen

The #1 cause of wrinkles is sun damage, so its important to use a sunscreen of at least 30 SPF from your early years on even in winter and on cloudy days. A great trick is to purchase two moisturizers: One for night and one for day that includes UV protection. Dont use moisturizers with sunscreen at night, the ingredients are not meant to be used 27/7 and can aggravate skin. When choosing a sunscreen, make sure it contains Mexoryl (found in my favorite sunscreen La-Roche Posay) or Helioplex, found in Neutrogena products.

6 Step How to Increase Your Memory

Forget and remember are two things that most often experienced by men all her life. By considering all the important things we can certainly make things easier, otherwise if a little forget and forget again, so beware of people who go senile.

There are 6 ways you can help us increase the ability to remember. Hopefully the following therapies can help.

1. Wake up and smell the rosemary.
In a study of 2003, the psychologists asked 144 volunteers to conduct a series of trials of long-term memory, working memory means, and test the reaction and attention. Some people do the test in an odor-free room, some in the room with the smell of rosemary essential oil, and the rest worked with husband smells of lavender.
blog-apa-aja.blogspot.com
The result, those working in the room by having great results in long-term memory and working memory compared with working in a room without smells, while working in the lavender-scented room worse in terms of working memory. Furthermore, those working in the rosemary-scented room feel more awake than those who work in the control room (no smell). Well, who worked in the lavender-scented room was more to feel sleepy.

Well, apparently its good lavender to repel mosquitoes. But side effects can make us prefer to sleep.

2. Food Boost Your Memory
To keep the memories remain young despite the aging brain, researchers suggest that eating foods rich in antioxidants like blueberries, apples, bananas, dark green vegetables, onions, and carrots.

blog-apa-aja.blogspot.com
Antioxidants are molecules that bind and neutralize easily electrons are called "free radicals" that roam freely in the bloodstream. These free radicals increases with age and can kill brain.

Secondly, most of the brain is made of healthy fats, including the most important are the omega-3. So that the brain can repair itself and become neurons are connected properly, we must provide appropriate food for the brain. Well, omega-3 found in many species of fish and nuts.

3. Chewing gum
Research conducted in 2002 in England found that chewing gum providing better results in long-term memory test and short-term than those who did not chew anything.
blog-apa-aja.blogspot.com

The scientists suspect, the act of chewing gum will produce saliva, which increases heart rate, or it affects the functioning of the brain region called the hippocampus that causes the body to release insulin in preparation for the metabolism of food.

4. Brain Game
A program called Lumosity, developed with the help of neuroscientists and cognitive psychologists from Stanford University and the University of California at San Francisco, is specifically designed for parents who want to improve memory, concentration, waking, and even their mood.
blog-apa-aja.blogspot.com
Of course, theres always sports a classic brain and cheap, such as Sudoku and crossword puzzles that can be found anywhere. Exercises that will inspire knowledge and help the nerves in the brain interconnected.

5. Sleep.
In a study on lab scale using rats, while rats slept, two areas in the brain - the hippocampus and medial prefrontal cortex, an area associated with memory retrieval of the past (either in human or rat) - rotating display events throughout the day. This process is believed essential to consolidate and tidy up the files of new memories are formed.
blog-apa-aja.blogspot.com

6. Walk
Research shows that the memory center in the brain called the hippocampus shrinks with age. But research in 2011 to give good news: people grow older adults who regularly walk to maintain the volume of the hippocampus.
blog-apa-aja.blogspot.com

The study, led by Arthur Kramer of the University of Illinois Champaign-Urbana that involved 60 adults aged 55 to 80 years. They did walk three times a week each for 40 minutes.

Enough activity to increase their heart rate. Other participants are the same number of muscle toning exercises weight training, yoga, and stretching, the same intensity.

After a year of tightening, the anterior hippocampus of participants lost a little over 1 percent of its volume, on average. In contrast, aerobic exercise a year makes about 2 percent increase in volume of the anterior hippocampus, reversing the natural aging hippocampus for about two years.

Scientists believe that it is caused by exercise-induced mild stress that triggers the production of growth factors in the brain. Possibly also due to greater blood flow to the brain so that more nutrients and oxygen are produced.

Healthy Habit No 8 Protect Your Skin

Our skin starts to age as soon as we are born and, according to the American Academy of Dermatology (AAD), the best way to protect it and look younger is to stay out of the sun.

The sun has harmful ultraviolet (UV) rays that can cause wrinkles, dryness, and age spots. Overexposure can cause sunburn, skin texture changes, dilated blood vessels, and skin cancers.

Avoiding the sun, however, is not always ideal or practical. To reduce the risk of skin damage, the AAD offers the following tips:

* Always wear sunscreen with SPF 15 or higher.
* Don a hat with a brim and wear other protective clothing.
* Dont deliberately sunbathe.
* Try to avoid sun exposure between 10 a.m. and 3 p.m.

Inflammation silent killer Learn how it can affect your health

How do you know if you have inflammation?

11 symptoms associated with low-grade chronic inflammation.
• body aches and pains
• congestion
• frequent infections
• diarrhea
• dry eyes
• indigestion
• shortness of breath
• skin outbreaks
• swelling
• stiffness
• weight gain/obesity

One of the main reason you put on more weight when you are under  stress is due to a hormone in your body called cortisol.

Cortisol is released in much higher quantities than normal when you are stressed out, which increases your blood pressure and blood sugar level. This can be a major contributor to you gaining weight.

People who are stressed out are not likely to lose weight.  We  dont feel this inflammation yet it gradually damages the ability to gain energy from food consumed. Over time, the inflammation promotes diseases regarding the circulatory system and type 2 diabetes.

Inflammation makes people fat by eating creating wear on the cells causing the cells to become inflamed. When the cells are inflamed, they do not use insulin correctly resulting in weight gain and fat.

The body has natural enzymes that neutralize the free radicals, AGE’s, as well as other damaging substances in the body. The diet rich with processed food damages the body’s natural abilities to remove the hazardous substances leading to the inflammation of the cells and arterioles.

This makes the enzymes fight harder to remove the substances reducing the resistance levels of the body. The body becomes overwhelmed causing people to experience oxidative stress thus leading to the inflammation and weight gain.

To help reduce the possibilities of inflammation reduce the amounts of processed foods consumed. Increase the natural foods of vegetables, lean meats, and fruits. A healthy diet will reduce the changes of the inflammation and excessive weight gain. Diets rich in natural whole grains, fruits, and vegetables also helps to promote weight loss by removing the processed foods that lead the body to storing additional fat and other damaging substances.

Dozens of pharmaceutical drugs have been developed to override the inflammatory cascade, and many more are in the pipeline. NSAIDs (nonsteroidal anti-inflammatory drugs) like Motrin and Aleve disrupt the production of prostaglandins, which are needed to regulate inflammation, constrict or dilate vessels, and much more. Corticosteroids like prednisone, COX-2 inhibitors like Vioxx and Celebrex, and antihistamines each shut down a different inflammatory mechanism, leading to further long-term risks in the body.

The mangosteen fruit is a powerful natural anti inflammatory. A natural solution without negative side effects. Mangosteens are grown in Southeast Asia and have been used for centuries in healing.

Personally I drink Xango 3 times a day!  Its a powerful known anti inflammatory that got me off of ibrupofen a few years ago I wont go a day without it.  Xango is the only mangosteen juice on the market that uses the whole fruit puree, rind and pericarp of the mangosteen which has xanthones with science and research to back it up.  Learn more about Xango here.
